Expression of Glutathione transferase isoenzymes in human hepatoma tissues

Abstract

The activity and isoenzyme pattern if glutathione tranafersea(GST) was compared immunochemically between human liver cancers and the correspondinfg pathologically cancer-free resection margin tissues. Total GST activity toward chloro-dinitrbenzene(CDNB) as a subatrate was decreased in cancer bearing parts than in cencer-free pats. In order to study the isonzyme pattern of GST in cancer tissues, the basci forms of GST isoenzymes were purified from rat liver tissues through DEAF-cellulose column and affinity chromatography. The polyclonal antibody rased aginst the basic isoenzymes of GST was not immuncross-reactive the acidic GST from the placental tissues(GST-n)and vice versa. Thereforce, it was possible to illustrate the differential exprssion of GST isoenzymes in the tissues with the respective antibody. Western blot analysis showed that antibodies raised aginst GST-n could detect thr protein of 25 kDa in cytosols of both the cancer bearing and the cancer free tissues, but significantly hugher level in thr part, while the protein detected by antibody against rat liver basic GSTs(GST-L), was decreased in the corresponding cancer bearing part. Immunochistochemical stuies also confirmed that acidic GST was cheifly expressed at cancer-bearing regions where the expression of basic GSTs were almost negligible, Moreover, the bile dutules were strongly positrve in immunistochemical staning with anti GST-n antiboy.
